Experimental Methods:
SeekOne™ DD Single-Cell 3’ Transcriptome-seq Kit was used to perform single-cell separation, nucleic acid labeling, and capture on the SeekOne™ Digital Droplet System. The high-throughput single-cell transcriptome libraries were sequenced using the Illumina NovaSeq 6000 platform.
Data Analysis:
Raw sequencing data was converted to fastq format with Bcl2fastq, then low-quality data was filtered out using the fastp quality filtering software. The filtered sequencing results were analyzed with the SeekSoul Tools, generating intermediate files like single-cell library quality control results, expression matrices, and alignment results. SeekSoul Tools supports counting reads that map to intron regions for UMI counting, which is more suitable for nuclei isolated samples.
